Anuj Dal will serve as his vice-captain. The allrounder has scored 254 runs and taken two wickets in 19 50-over appearances.
Derbyshire finished seventh in Group B in 2022, winning three of their eight matches, but haven't reached the knockout stages of the 50-over competition since 2014.
Head of cricket, Mickey Arthur, said: "Brooke has shown his ability to contribute to the team effort and make a difference in games on a number of occasions this season and now he has the opportunity to lead our team in the One-Day Cup.
"He will be supported by Anuj, who is a key member of our dressing room and someone who is eager to make a difference as a senior player, as well as a few experienced heads within the squad."
