Matches will take place at the National Stadium until February 7 before the tournament relocates to Lahore.
Quetta are due to get their campaign under way against Peshawar Zalmi on Friday, and are due to play five matches in Karachi.
Afridi, meanwhile, will need to submit a negative test to return to the Gladiators camp at the end of his quarantine period.
Quetta general manager Azam Khan was quoted by Pakistani media as saying that the allrounder is asymptomatic.
